The systems shears RNA and DNA molecules for next generation sequencing library preparation.The MRC -Universlty of Glasgow Centre for Virus Research (CVR), Institute of Infection, Immunity and Inflammation, needs a focused ultrasonicator that can do 96_well plates. Focused ultrasonication of DNA is the method of choice for fragmenting nucleic acid because it is unbiased and reproducible, independently of the amount of starting material.Mandatory Technical Requirements of the system:1) Reproducibility: Acoustic shearing of DNA, produces fragments in a highly reproducible size range down to 100 bp, in contrast to other techniques such as nebulization with nitrogen gas, sonication, and hydroshearing.2) Capability to process low sample volume and mass: fragmentation in low volumes (as low as 15 ul) overcomes the need to re-concentrate fragmented samples, a process typically associated with sample loss. Additionally, many of the samples processed are low input and other methods such as enzymatic fragmentation are highly irreproducible.3) Unbiased fragmentation: it is essential to minimize bias introduction during the library preparation.4) Low risk of sample contamination and mix up: there should be no physical contact between the , instrument and the sample DNA to prevent sample to sample contamination. This is an essential criteria in NGS.5) Low hand on time: Automated batch processing from 1 up to 96 samples is an essential feature when doing high throughput sequencing.6) Safety: the system must operate outside the human audible range so the equipment can be operated in a general laboratory bench setting without the extra expense of system enclosures to ensure proper hearing protection.CVR department requires a benchtop instrument only, with a footprint of not more than 65cm x 80cm. Space in the CVR is very limited, and a larger instrument will be difficult to accommodate.The only system that meets all mandatory criteria and supports technical functionality needs is the COVARIS LE220 focused-ultrasonicator.Purchasing Policy Exception form with full details has been completed to support this requirement.
